Abstract

The bougenville orange (Bougainvillea x buttiana) has been reported to have antioxidant activity using the DPPH method. Testing antioxidant activity can be carried out using various methods. In this study, the antioxidant activity of orange bougenlille (Bougainvillea x buttiana) flower was tested using three methods, namely the DPPH (2,2-diphenyl-1-picrylhydrazyl), FRAP (Ferric Reducing Antioxidant Power) method and the FTC (Ferric Thiocyanate) method. The research began with extraction using methanol and extraction using solvents with increased polarity (n-hexane, ethyl acetate, methanol). Each extract and fraction were determined the total phenolic and flavonoid content and antioxidant activity using the DPPH, FRAP and FTC methods. The results showed that the highest levels of total phenolics and flavonoids were found in the ethyl acetate fraction, 35.51GAE/g and 25.00 mgQE/g respectively. Antioxidant activity using the DPPH and FRAP methods showed the highest activity in the ethyl acetate fraction with an IC50 of 18.59 mg/L for the DPPH method and reducing power with an Fe2+ concentration of 115.75 mg/L with the FRAP method. The antioxidant activity using the FTC method showed the highest activity in the total methanol extract with % reduction of 45.16%. Based on the data obtained, the antioxidant activity of B. x buttiana flower using the DPPH and FRAP methods was more effective at the ethyl acetate fraction. The results obtained are comparable to the phenolic and flavonoid content, whereas in the FTC method the total methanol extract was more effective than fractions. This is indicated that polar compounds have a good ability to inhibit the formation of fatty acid peroxidation.
