Abstract
The objective of this research was to isolation bacteria that produced protease and know characteristics of protease enzyrae from swamp mud bacteria at Sakatiga Indralaya, Ogan Ilir, South Sumatra. The research was conducted from July until August 2008 in Technology of Fishery Product Laboratory, Aquaculture Laboratory, Agricultural Faculty and Microbiology Laboratory Mathematics and Natural Sciences Faculty, Sriwijaya University. The research used laboratory method that share of some in stage begin of screening microbial that production protease from microbial or bacteria of swamp mud at Sakatiga Indralaya until production bacteria that has protease. The result showed that isolate bacterial from swamp mud has produced 3 isolate bacterial with has that contain protease that are TjSi, T3S1 and T3S2. The optimum activity in production of protease happen at hour. Which has the activity value protease at isolate T1S1 has 0,318 IU/ml, isolate T3S1 has 0,391 IU/ml and isolate T3S2 0,385 IU/ml. The optimum pH at isolate T1S1 was 7.5, while to T3S1 and T3S2 was 8. The optimum temperature at isolate T1S1 was 50 °C, isolate T3S1 was 40 °C and isolate T3S2 was 50 °C. Metal ions FeCfe (1 mm) increased enzyme activity at isolate T1S1 and T3S1. KC1 (1 mm) gave an inhibitory effect. Metal ions at isolate T3S2 had no effects to the enzyme.
