Abstract
The aim of this research was to obtain the best callus formation in vitro culture by addition of 2,4-D (2,4-dicholorophenoxyacetic acid) and picloram (4-amino-3,5,6-tricloropicolinic acid) to induced duku’s leaf explants (Lansium domesticum Corr.). This research was conducted from February 2014 to October 2014 at Tissue Cultrue Laboratory, Department of Agronomy, Faculty of Agriculture, Sriwijaya University, Ogan Ilir, Palembang, South Sumatera. The culture medium which was used WPM (woody plant medium) with 2.4-D and picloram treatments and each of them contained of 0 ppm (control), 2 ppm, 4 ppm, and 8 ppm. This research was arranged in completly randomized design with 7 treatments and 5 replications. Each unit of treatment included of 6 culture bottles (sub replication) so that the total were 210 culture bottles. The result showed that duku’s leaf explant could make callus by additon of 2,4-D and picloram. Additin of 2,4-D with 4 ppm concentration had show the best result. The colour of growth callus was white, yellow to brown and also had the crumb texture.
