Abstract

Breadfruit leaves (Artocarpus altilitis) are one of the herbal medicines that have activity as antihyperglycemia, antibacterial, anti-inflammatory, and antihypertensive. From the results of phytochemical screening, it is known that ethanol extract of breadfruit leaves contains phenolics, saponins, tannins, flavanoids, polyphenols, hydrocyanic acid, acetylcholine, riboflavin. The safety of using breadfruit leaves as a natural medicinal preparation must be supported by scientific research on toxicity testing. In this study, an acute toxicity test of ethanol extract of breadfruit leaves was carried out on female Wistar rats using the Acute Toxic Class Method. In this method, preliminary tests were carried out to obtain the initial dose used in the main test. Based on preliminary tests, a dose of 10,000 mg/kgBB was obtained as the initial dose for the main test. The results of observations in the main test group showed no deaths in the test animals which indicated that there was no toxic effect from EEDS. This is reinforced based on the examination of biochemical parameters obtained the results of no significant differences in the levels of SGOT, SGPT, ureum, LDL, and Trigriserida (p>0.05), but there is a significant difference in creatinine levels (p<0.05). The average levels of biochemical parameters in the normal control group were SGOT 111.96 ± 12.01 U/L, SGPT 44.98 ± 11.39 U/L, creatinine 0.37 ± 0.06 mg/dL, urea 29.12 ± 6.47 mg/dL, LDL 4.89 ± 1.46 and trigriserida 67.31 ± 10.05. While the treatment group was SGOT 111.36±15.69 U/L, SGPT 56.38±4.76 U/L, creatinine 0.43±0.01 mg/dL, ureum 23.71±4.82 mg/dL, LDL 5.28±1.18 and trigriserida 67.31±5.31. The main test group administered EEDS 10,000 mg/kgBB showed the effect of macroscopic liver damage indicated by fatty liver. The results of organ histopathology in the main test group showed reversible hydropic degeneration of liver and kidney organs.
