Abstract

The ethanol extract of Pelawan leaves (Tristaniopsis merguensis Griff.) contains flavonoids, steroids, phenolics, and terpenoids, which have the potential to act as analgesics and anti-inflammatory agents due to their ability to inhibit prostaglandin formation, the main mediator of pain and inflammation. This study aimed to evaluate the characteristics of the extract as well as the analgesic and anti-inflammatory effects of the ethanol extract and fractions with varying solvent polarities from Pelawan leaves. The evaluation was based on the reduction in writhing response in rats induced by 0.6% v/v acetic acid and the decrease in edema volume induced by 1% b/v carrageenan. The analgesic and anti-inflammatory tests were conducted using the writhing test and rat paw edema methods on 24 male Wistar rats, which were divided into six treatment groups. Group I, the negative control group, received 1% Na CMC, Group II, the positive control group, received Na diclofenac, while Groups III, IV, V, and VI served as test groups, all receiving 100 mg/kg BW with different solvent polarities: ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction. The analysis of compound composition in the extract is conducted using Gas Chromatography-Mass Spectrometry (GC-MS), which revealed five peaks. The compounds identified inclued β-Caryophyllene, α-humulene, β-selinene, Aromadendrene, dan Dodecanoic Acid. The analgesic and anti-inflammatory percentages for the positive control group were 75.5% and 86.05%, respectively. Groups III, IV, V, and VI demonstrated analgesic and anti-inflammatory percentages of 25.00% and 54.42%; 70% and 80.47%; 64.07% and 58.32%; 20.47% and 17.49%, respectively. These findings indicate that the n-hexane fraction was the most active and optimal as an analgesic and anti-inflammatory agent, with effects comparable to the positive control group treated with Na diclofenac (p>0.05).
