Abstract

Moringa leaf (Moringa oleifera L.) are proven to contain secondary metabolites, one of which is flavonoids in the form of quercetin which has the ability as an anticancer. This study aims to determine the optimum formula of nanoparticles from a variety of alginate chitosan coatings at concentrations of 0.3% and 0.6% conjugated with folic acid. Moringa leaf extract was obtained through maceration using ethanol with a yield percentage of 17.58%. FTIR characterization of chitosan folic acid conjugate showed successful conjugation with 99% efficiency of successfully conjugated folic acid. Optimization using 2² factorial design method with Design Expert® 13 resulted in an optimum formula at 0.3% chitosan and 0.6% alginate, with a particle size of 133.6 nm, PDI 0.272, zeta potential -27.6 mV. The characterization results of the optimum formula showed a decrease in encapsulation efficiency from 92.216% on day 0 to 81.267% on day 12, as well as a pH reduction from 7.43 to 7.37 after the cycling test. Moringa leaf extract nanoparticles can still be categorized as stable, supported by good visual stability and the ability to withstand gastrointestinal pH (pH 7.4) with a log k value of -0.7985. TEM showed the chitosan-alginate gel structure as evidence of successful conjugation and drug release reached 70% in 24 hours which followed the Hixson Crowell release model. The optimum formula of moringa leaf extract nanoparticles showed good characterization and stability, with chitosan as the main carrier of effective folic acid. Moringa leaf extract nanoparticles formulated with chitosan as a folic acid carrier show great potential as a stable and effective drug delivery system, which can improve the efficacy of anticancer therapy by controlling targeted drug release and minimizing side effects on healthy tissues.
