Abstract

Ofloxacine is a fluoroquinolone antibiotic that is classified under Biopharmaceutics Classification System (BCS) Class II, characterized by low water solubility and high permeability. This investigation aimed to enhance its solubility by utilizing a combination of sodium alginate and chitosan as carrier polymers in a solid dispersion system. The development of the optimal formulation for ofloxacin solid dispersion was carried out by observing variations in the amounts of chitosan and sodium alginate. The optimization process was conducted using Design-Expert® software with a 2² factorial design method. The solid dispersions were prepared using the co-grinding technique, and the optimum formula was identified based on the highest desirability value and the best characterization results, including dissolution rate, flow rate, angle of repose, compressibility index, and Rasio Hausner. The formulation design consists of four formulas, namely F1, F2, F3, and F4, each containing 50 mg of ofloxacin with varying amounts of sodium alginate (80–100 mg) and chitosan (70–100 mg). The optimization results indicate that F2 is the optimum formula, containing 70 mg of chitosan and 100 mg of sodium alginate. Characterization of the optimum formula demonstrated a 14.235% reduction in crystallinity through XRD analysis, there is no interaction was found between the carrier polymer and ofloxacin in the FTIR test, and a morphological transformation from crystaline phase into an amorphous phase observed through SEM analysis. The solubility of the optimum formula increased significantly in SGF solution. Based on the findings, the optimal formulation of ofloxacin solid dispersion has demonstrated good characterization outcomes and the ability to enhance the solubility and bioavailability of ofloxacin.
