Abstract

Kanker payudara merupakan penyebab utama kematian pada wanita, sehingga diperlukan agen terapeutik baru yang lebih efektif dan minim efek samping. Penelitian ini bertujuan mengeksplorasi komponen bioaktif jaras gambir sebagai agen antikanker payudara, menganalisis interaksinya dengan protein target melalui network pharmacology, serta mengevaluasi mekanisme molekulernya menggunakan molecular docking. Network pharmacology digunakan untuk mengidentifikasi target protein dan mekanisme molekularnya, sedangkan molecular docking mengevaluasi interaksi senyawa aktif dengan protein target. Hasil penelitian menunjukkan bahwa senyawa dalam jaras gambir memiliki afinitas kuat terhadap protein yang berperan dalam proliferasi dan apoptosis sel kanker, seperti AKT1, TP53, BCL2, TNF, dan EGFR. Interaksi ini dikonfirmasi melalui binding affinity serta ikatan hidrogen dan hidrofobik pada residu kunci. Jalur utama yang terlibat meliputi PI3K-AKT, p53 signaling pathway, dan TNF signaling pathway. Senyawa seperti chlorogenic acid dan isoquercitrin berinteraksi dengan target protein melalui ikatan hidrogen dan hidrofobik, sementara morin hydrate, naringenin, quercetin, eriodictyol, ribofuranoside dan scopoletin menunjukkan aktivitas serupa dengan obat pembanding (erlotinib). Hasil ini menegaskan bahwa delapan senyawa tersebut menjadi senyawa terbaik yang terkandung pada jaras gambir dengan potensi sebagai agen terapi kanker payudara.
