Abstract
Mangosteen peel extract (Garcinia mangostana L.) has alpha mangostin compounds that have very strong antioxidant activity. Transethosomes have better vesicle elasticity and penetration levels so that they can increase the effectiveness of alpha mangostin. This study aims to determine the optimum formula of transethosomes from variations of 96% ethanol concentrations of 20% and 40% with tween 80 concentrations of 0.5% and 0.75%. Optimization using the 22 factorial design method with the Design Expert® 13 application with responses of %EE, decrease in %EE, pH, decrease in pH, and viscosity. The optimum formula of transethosomes was characterized, tested for transethosomes morphology, and tested for antioxidant activity. Based on the results of the Design Expert® 13 analysis, optimum formula was found in formula 2 with concentration of 20% ethanol and 0.5% tween 80 with a %EE value of 99.480 ± 0.2626, a decrease in %EE of 0.362 ± 0.1010, pH 7.62 ± 0.1501, a decrease in pH of 0.030 ± 0.0137, and a viscosity of 1.377 ± 0.4980 cP. The characterization results of the optimum transethosome formula obtained a particle size of 225.9 ± 2.193 nm, PDI 0.28 ± 0.026, and a zeta potential of -8.00 ± 0.695 mV. Transethosome has a spherical shape and the purified mangosteen peel extract transethosome 90% alpha mangostin obtained an IC50 value of 70.91 ppm. Based on the results obtained, the optimum formula of transethosome purified mangosteen peel extract 90% alpha mangostin has good characterization results and has strong antioxidant activity.
