Abstract

This study was aimed to evaluate the response of young leaf explants of oil palm from different ortet ages and determine the most responsive ortet age. This research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University, Palembang City, South Sumatra. The research was conducted from November 2024 to February 2025. This study used a completely randomized design (CRD) method. Variations of ortet ages were set as treatments consisting of ortet with ages of 8 months (P1), 10 months (P2), and 12 months (P3) which were repeated four times. Each unit consisted of 10 experimental bottles. Explants were planted on Murashige and Skoog media with addition of Naphtalene Acetic Acid (NAA) 6 ppm and 2,4-Dichlorophenoxyacetic acid (2,4-D) 0,5 ppm. The results showed that the highest of explant contamination occurred in explants from 8-month-old ortets (20%), followed by the 10-month treatment (12,5%) while the 12-month treatment had the lowest contamination (7,5%). The level of explant contamination affected the percentage of explant life, the explants from 12 months old ortet had the highest percentage of live explants (92,5%), followed by 10 months (87,5%) and 10 months (80%) treatments. At 12 weeks after inoculation (WAI), the explants did not show any callus formation on the explants, but many explants showed swelling as a sign of callus induction, and swelling occurred at five weeks after inoculation (WAI). The highest percentage of swollen explants was in the 12-month treatment (67,5%), followed by the 10-month treatment (52,5%) and the 8-month treatment (35%). It is recommended to use explants with an age of ≥12 months to increase the success and efficiency of tissue culture.
