Abstract
This study was aimed to evaluate the growth among different treatment combinations of NAA and explant size to get the best treatment combination for initiation of callus from young leaves of oil palm (Elaeis Guineensis Jacq.). The research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Sriwijaya University, Bukit Lama, Ilir Barat I District, Palembang City, South Sumatra, at coordinates 2°59’23.4”S; 104°43’53.4”E. Research was carried out from November 2024 to February 2025. The research design was Completely Randomized Design (CRD) with two factors, the treatments were NAA and explant size, NAA concentration : 1 ppm (N1), 3 ppm (N2), and 6 ppm (N3) and explant size consisted of 0.5 cm (S1), 0.75 cm (S2), 1.0 cm (S3), 1.25 cm (S4), and 1.5 cm (S5) with four replications, and three culture bottles per unit. Based on the analysis of variance of NAA concentration and explant size, the treatments had significant effects on explant contamination, live explants and stagnant explants and had no significant effect on other variables. The combination of 3 ppm NAA with explant size of 1 cm (N2S3) produced the most explant swelling with a percentage around 75%. The growth of explant producing callus did not occur until 12 weeks after inoculation. Keywords: Explant Size, Oil Palm, Naphthalene Acetic Acid (NAA).
