Abstract

Introduction. The ongoing emergence of drug-resistant tuberculosis (DR-TB) cases globally poses challenges for comprehensive management. Linezolid, used in DR-TB treatment, is associated with adverse effects such as toxic optic neuropathy (TON). The occurrence of TON has been linked to mitochondrial DNA (mtDNA)polymorphisms. This study aimed to determine the association between the G3010A mtDNA polymorphism and the incidence of TON in DR-TB patients receiving linezolid. Methods. This was an observational analytic study with a cross-sectional design, conducted from January to March 2025. A total of 51 DR-TB patients from the Integrated DR-TB Clinic at Mohammad Hoesin General Hospital were included. Blood samples were collected for G3010A mtDNA polymorphism testing using PCR-RFLP at the Biotechnology Laboratory, Faculty of Medicine, Universitas Sriwijaya. Results. Most patients were male (62.7%), aged 30–60 years (60.8%), without diabetes mellitus (78.4%) or HIV (94.1%). The majority had MDR-TB (50.1%) and were treated with individual regimens (62.7%). The incidence of TON among patients receiving linezolid was 43.1%. No G3010A mtDNA polymorphisms were detected in any of the study subjects. Conclusion. The association between the G3010A mtDNA polymorphism and TON in DR-TB patients receiving linezolid could not be determined in this study. Further investigation is needed to identify other potential risk factors for TON in this population.
