Abstract

Holothuria atra, Acanthaster planci, and Culcita novaeguineae as part of the Echinodermata phylum have the potential as a source of natural collagen that can be utilized in pharmacology. Collagen has potential bioactivities that act as antiaging, antioxidant, anti-inflammatory, and antimicrobial. This study aims to isolate and characterize collagen from the Echinodermata phylum in revealing the potential of marine natural products. Samples were taken from the neritic zone of Pahawang Island, Lampung. Antioxidant activity assessed through DPPH assay showed very strong activity (IC50 < 50 µg/ml) for all samples, with IC50 values of 10.84 µg/ml (H. atra), 21.78 µg/ml (A. planci), and 4.43 µg/ml (C. novaeguineae). LC-MS analysis identified major amino acids, including Glutamic acid, Aspartic acid, Valine, Leucine, Lysine, Isoieucine, Alanine, Glycine, Proline, Threonine, Serine, Methionine, Phenylalanine, Histidine, and Arginine, which were confirmed the existence of their compound structures by ¹³C carbon NMR signal analysis. FTIR spectroscopy detected typical collagen functional groups, including Amides I, II, III, and A. The results of molecular docking of Glutamic acid ligand (B:GLU921) to NMDA receptor (PDB:4PE5) yielded an affinity energy (ΔG) of - 5.25 kcal/mol and a Ki value of 142.02 μm, indicating its role as an endogenous agonist and potential NMDA receptor modulator. These findings provide important information in the disclosure and development of marine-derived natural products for therapeutic applications.
