Abstract

Nile tilapia (Oreochromis niloticus) is one of the freshwater fish species in the Cichlidae family with several strains that have been genetically selected and developed, including red tilapia, GIFT, and BEST. The purpose of this study was to determine the percentage of species identity, genetic distance, and phylogenetics of Nile tilapia from Lake Ranau, both from nature and culture, with species in the GenBank data center. This research was conducted in the Plant Physiology Laboratory and the Biotechnology Laboratory, Agronomy Study Program, Faculty of Agriculture, Universitas Sriwijaya, in December 2024-January 2025. The pectoral fins of eight Nile tilapia were used, consisting of, 4 samples of wild, (TRA) and four samples of culture (TRB) from Lake Ranau. The study was conducted in several stages, namely DNA extraction, PCR (Polymerase Chain Reaction) amplification, electrophoresis, DNA visualization, and cyt b gene sequencing. The mtDNA cyt b gene fragment was amplified at 49.7°C for 30 seconds with 32 cycles. The nucleotide length of the cyt b gene is 739 base pairs (bp). BLASTn (Basic Alignment Search Tool Nucleotide) results indicated that TRA2, TRA3, TRA4, and TRB4 had the highest similarity (99.87-100%) to O. niloticus from China and Nigeria. Samples TRA1, TRB1, and TRB5 had the highest similarity (100%) with Oreochromis sp. from China. The genetic distance of all samples, both natural and culture, had a value of 0.0%. Phylogenetic tree construction showed that all samples were grouped in two main clusters. The first cluster consisted of TRA2, TRB4, TRA3, TRA4, while the second cluster consisted of TRB1, TRB5, and TRA1.
