Abstract
Skin diseases are usually caused by microbes that infect the skin. Staphylococcus aureus bacteria are one of the bacteria that can cause diseases on the surface of the skin and nose of humans. Other microbes that can infect the skin are Candida albicans. The use of chemical drugs to treat skin infections repeatedly can cause skin-infecting microbes to become resistant to the drugs given, therefore research is needed on alternative treatments that do not cause resistance from microbes. The ketapang plant (Terminalia cAtappa L.) is often used by Asian people as a traditional medicine to treat skin diseases such as rashes and itching. Ketapang leaf extract is known to have antifungal, antibacterial and anti-inflammatory properties. This research was conducted from September 2024 to May 2025. This research was conducted at the Microbiology Laboratory, Genetics and Biotechnology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. The results of the study showed that the secondary metabolite extract produced from endophytic fungi of Ketapang plants (Terminalia cetappa L.) has antimicrobial potential as evidenced by the formation of inhibition zones and groups of secondary metabolite compounds produced by endophytic fungi of Ketapang plants (Terminalia catappa L.) including tannins, flavonoid and Terpenoid. The minimum inhibitory concentration (MIC) indicated also has a low value of 21.25 ppm against Staphylococcus aureus and 11.25 ppm against Candida albicans, thus having greater potential for its application. The endophytic fungi identified in this study have antimicrobial capabilities against Staphylococcus aureus and Candida albicans with strong criteria, namely Cladophialophora, Neurospora, Cladosporium, and Umbelopsis. Keywords: Ketapang, Staphylococcus aureus, Candida albicans, endophytic fungi and Penyakit kulit biasanya disebabkan oleh mikroba yang menginfeksi kulit. Bakteri Staphylococcus aureus merupakan salah satu bakteri yang dapat menyebabkan penyakit pada permukaan kulit dan hidung manusia. Mikroba lainnya yang dapat menginfeksi kulit yaitu Candida albicans. Penggunaan obat kimia untuk mengatasi infeksi kulit secara berulang dapat menyebabkan mikroba penginfeksi kulit mengalami resisten terhadap obat yang diberikan, karena itu diperlukan penelitian mengenai alternatif pengobatan yang tidak menimbulkan resistensi dari mikroba. Tanaman ketapang (Terminalia catappa L.) sering digunakan oleh masyarakat Asia sebagai obat tradisional untuk mengatasi penyakit kulit seperti ruam dan gatal-gatal. Ekstrak daun ketapang diketahui memiliki sifat antifungi, anti bakteri dan anti inflamasi. Penelitian ini dilaksanakan pada bulan September 2024 sampai dengan Mei 2025. Penelitian ini dilakukan di Laboratorium Mikrobiologi, Laboratorium Genetika dan Bioteknologi Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam, Universitas Sriwijaya. Hasil penelitian menunjukan bahwa ektrak metabolit skunder yang dihasilkan dari fungi endofit tumbuhan Ketapang (Terminalia catappa L.) memiliki potensi antimikroba yang dibuktikan dengan terbentuknya zona hambat dan golongan senyawa metabolit sekunder yang dihasilkan oleh fungi endofit tumbuhan Ketapang (Terminalia catappa L.) diantaranya yaitu tanin, flavonoid dan Terpenoid. Konsentrasi hambat minimum (KHM) yang ditunjukkan juga memiliki nilai yang kecil dengan konsentrasi 21,25 ppm terhadap Staphylococcus aureus dan 11,25 ppm terhadap Candida albicans, sehingga memiliki potensi yang lebih besar dalam pemanfaatannya. Fungi endofit pada penelitian ini yang teridentifikasi memiliki kemampuan antimikroba terhadap Staphylococcus aureus dan Candida albicans dengan keriteria kuat yaitu Cladophialophora, Neurospora, Cladosporium dan Umbelopsis.
