Abstract
Thrombosis can trigger diseases such as ischemic stroke and acute myocardial infarction. Thrombosis therapy is usually administered with antiplatelets, anticoagulants, and thrombolytic agents. Unlike antiplatelets and anticoagulants which are preventive, thrombolytic agents work directly to dissolve thrombus, for example tPA. Tempe bongkrek is a food fermented by Rhizopus oligosporus fungus that contains proteolytic enzymes. The purified enzyme of tempe bongkrek that has been studied has fibrinolytic activity. This study aims to determine the optimum formula of liposome photothermal sensitizer that encapsulates crude extract and purified enzyme of tempe bongkrek. Determination of the optimum formula using Design-Expert® with factorial method 22. The factors used are soy lecithin and tween-80. The maximum and minimum levels used for soy lecithin were 0.8% and 0.1% and for tween-80 were 0.1% and 0.01%. The optimum formula obtained was characterized by particle size, PDI, morphology, zeta potential, cycling test (consisting of physical appearance test and encapsulation efficiency), thrombolytic activity and photothermal efficiency. The concentration of soy lecithin can increase %EE, viscosity, and temperature increase but soy lecithin can reduce pH and Tween-80 can reduce %EE, temperature increase and viscosity but has no significant effect on pH. The optimum formula obtained was 0.604% soy lecithin and 0.1% tween-80. Liposome photothermal sensitizer has a spherical shape with a particle size of 825.8 nm, PDI of 0.609 and zeta potential of -77.5 mV. Crude extract and purified enzyme of tempe bongkrek with 100% concentration had thrombolytic activity of 45.05% and 48.96%. The formulated liposome photothermal sensitizers demonstrated potential as thrombolytic agents, exhibiting thrombolytic activity with Lip-Phy-CETB, Lip-Phy-PETB, Lip-Flu-CETB, and Lip-Flu-PETB showing values of 32.40%, 41.98%, 16.00%, and 35.53%, respectively.
