Abstract

Latar Belakang: Thyroid eye disease (TED) merupakan gangguan inflamasi autoimun pada mata. Transforming Growth Factor Beta (TGF-β) mengakibatkan inflamasi pada otot ekstraokuler TED. Tujuan penelitian untuk menganalisis perubahan ekspresi TGF-β dan ketebalan otot ekstraokular setelah induksi oxazolone pada mencit model inflamasi orbita sebagai fitur klinis dari TED. Metode: Penelitian quasi eksperimental dengan desain rancangan runtut-waktu dilakukan pada Mencit BALB/c (Mus Musculus L) dengan fitur klinis TED yang di induksi Oxazolone di animal house dan laboratorium iRATco, Bogor. Sampel dibagi menjadi 6 kelompok (6 ekor per kelompok), yaitu K normal, K negatif, K1, K2, K3, K4 (diinduksi oxazolone, 3, 7, 14, dan 28 hari). Analisis data dilakukan dengan program SPSS versi 24. Signifikansi statistik P< 0,05. Hasil Penelitian: Sebanyak 36 ekor sampel diteliti. Didapati perbedaan ekspresi TGF-β pada serum darah, ekspresi TGF-β pada jaringan otot ekstraokuler antar kelompok interval waktu penelitian (nilai p = 0.013, p = 0.000, p = 0.000). Pada serum darah didapati kadar TGF-β antar kelompok interval waktu penelitian berbeda secara signifikan (nilai p = 0.000, p = 0.000, p = 0.027, p = 0.000, p = 0.000). Ekspresi TGF-β pada serum darah dan ketebalan otot ekstraokular memiliki korelasi sedang antar kelompok (r = 0.440, p = 0.000). Ekspresi TGF-β pada jaringan otot memiliki korelasi yang kuat antar kelompok (r = 0.638, p = 0.000). Simpulan: Ekspresi TGF-β pada serum darah dan ketebalan otot ekstraokular berkorelasi sedang dan kuat antar kelompok perlakuan.
