Abstract

Preparation and characterization of submicroparticles poly (lactic co-glycolic acid) (PLGA) loading methylprednisolone with polyvinyl alcohol (PVA) as stabilizer will make methylprednisolone more effective and stable since it can decrease side effect of drugs. PLGA particles loading methylprednisolone were prepared by using emulsion solvent evaporation method. There were three formulas (F) which were evaluated in this study based on the differences of sonication time, 5 minute for F1, 10 minute for F2, and 15 minute for F3. Results from measurement of percent encapsulation efficiency (% EE) using UV-Vis spectrophotometer were 5283.853 ± 0.038 % for F1, 85.580 ± 0.016 % for F2 dan 83.885 ± 0.043 % for F3. Characterization using particle size analyzer (PSA) on F2 resulted into 105.2 ± 31.22 nm with PDI of 0.621, which indicate homogeneously of partikel size was not good. The result of zeta potential measurement in F2 was -15.6 mV, which indicate that the stability of suspension was not good. Analysis of statistical parametric one-way ANOVA, post hoc Tukey and LSD showed that the difference of sonication time affected %EE for each formulas with p-value of <0.05. Based on the result of independent t-test with p-value of >0.05 for F1 and F2 showed that distribution of particle size for sample evaluation were not effected by differentiation of sonication time. Particle penetration testing has been done through in vitro diffusion test, using the dialysis membrane. The diffusion test showed the highest diffusion percent value on submicro particles of PLGAmethylprednisolon (85.896% ± 0.011) compared with pure methylprednisolone (30.695% ± 0.0001), and comercial methylprednisolone tablet dosage form (44.013% ± 0.001). Stability of dosage form was tested through heating cooling cycle method showed a decrease of insignificant level in each cycle.
