Abstract

Gambir leaf (Uncaria gambir Roxb.) contains flavonoid compounds, tannins, and alkaloids that can help the healing process of burns. Catechin as one of the main components in gambir leaf extract has function as anti-inflammatory, antibacterial, and antioxidant. Gambir leaf extract was formulated into spray gel to reduce the possibility of contamination or infection and pain when applied to the wound. Spray gel was formulated by using Carbopol®940 and HPMC-60SH as a gelling agent. In this study, 4 spray gel formulations were prepared with the ratio of Carbopol®940 : HPMC-60SH concentration i.e.: F1 (1% : 1%), F2 (5% : 5%), F3 (1% : 5%), and F4 (5% : 1%). Spray gel formulas were evaluated for physical properties such as organoleptic, homogeneity, pH, viscosity, adhesibility, and spray pattern. The results obtained that from the four formulas were only F1 and F3 which can be sprayed. Factorial design analysis showed that Carbopol®940 and HPMC-60SH factors had significant effect (p <0.05) on decreasing viscosity and adhesbility but increased spraying diameter. Result of factorial design showed the optimum formula ratio of Carbopol®940 and HPMC-60SH was 2.59%: 1% respectively with desirability value of 0.514. The burn wound healing activity of optimum formula was tested through wound healing percentage on rat back skin. Statistical test results by using Mann-Whitney showed wound healing effect did not differ significantly with positive control (p >0.05) during 14 days observation. It indicated that spray gel has same effect with positive control. Optimum spray gel gambir leaf extract has a burn wound healing effect as 56.20% at inflammatory and early stage of proliferation phase. Spray gel containing gambir leaf extract was able to accelerate healing process for burn wounds.
