Abstract

Diabetes mellitus (DM) type 2 is a metabolic disorder caused by cell resistance to insulin resulting in an increase in blood sugar levels. One of type 2 diabetes mellitus therapy is by inhibiting the activity of enzyme α-glucosidase. Kabau plant (Archidendron bubalinum (Jack) I.C Nielsen) is one of the plants traditionally used as an antidiabetic agent. The phenolic content in root and bark of kabau has potential as an inhibiting compound of α-glucosidase enzyme activity. This study aims to determine the effectiveness of antidiabetic of ethanolic extract of root and bark in kabau through inhibition test of α-glucosidase enzyme activity and determination of total phenolic content of extract. The result of ethanol extract test of root and bark in kabau showed inhibitor effect of α-glucosidase enzyme activity with IC50 value of each extract is 44,702 ppm and 65,665 ppm. The total phenolic content determined by spectrophotometry using Folin Ciocalteu. The total phenolic in ethanol extract of kabau root is 163 mg GAE/g and ethanol extract of kabau bark is 280 mg GAE/g. Based on the results, ethanol extract of kabau bark has the highest total phenolic content and inhibitory effect of α-glucosidase enzyme activity than kabau root.
