Abstract
The potential of thermo-lipolytic bacteria which can be utilized in various application in the industrial sector causes necessity research and exploration of local thermo-lipolytic bacteria. This research was conducted from November 2015 until completed in the Genetics and Biotechnology Laboratory, and Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Sciences, Sriwijaya University. This research was conducted to determine the isolates thermofilic bacteria which have the activity of lipolytic and to identification of thermo-lipolytic bacteria from the hot springs of Tanjung Sakti (South Sumatera) using gene encoding 16S rRNA. Stages of research conducted include thermo-lipolytic bacteria screening using pour plate method were performed using paperdisc on ZoBell agar medium enriched respectively with 1% olive oil, isolation of genomic DNA using DNA Wizard Purification Kit, amplification of 16S rRNA gene-based PCR, sequencing, and phylogenetic tree analysis using the MEGA6 application. The results of research, there are seven bacterial isolates that possess the activity of lipolytic, those are bacterial isolates TS1, TS2, TS4, TS5, TS12, TS14, and TS16. All of the isolates have therod-shaped cells and Grampositive. The result ofthe identification of those isolates areAnoxybacillus rupiensis TS1 and TS4 (99%), A. bogrovensis TS2 (86%), Brevibacillus thermoruber TS5 and TS14 (98% and 95%), Aeribacillus pallidus TS12 (99%), and Bacillus licheniformis TS16 (97%).
