Abstract
Coal mining is one source of state income and the source of environmental pollution through waste acid mine drainage ( AAT ) that it generates. AAT has a low pH and dissolved metals are high, so it will disrupt the aquatic ecosystem when AAT flows into the water body. Required remediation is effective, economical, and no produce second polutant to tackle the problem of AAT, using biological agents. L. flava able to live well and lower sulfate of 40.2 % in the bioreactor with AAT levels of 50 % allegedly cooperating with the bacteria in that rhizosphere. Therefore, it is necessary to do a research about isolation and identification of sulphate reducing bacteria from L. flava rhizosphere. The aims of the research is to get isolates of sulphate reducing bacteria from L. flava rhizosphere and knowing genera from that isolates. This research has been done on January 2016 to April 2016. The sampling was conducted in L. flava rhizosphere as bioremediation agent acid mine drainage from PT. Bukit Asam. The isolation, characterization, and identification was conducted in Microbiology Laboratory, Departement of Biology, Faculty of Mathematics and Natural, Sriwijaya University, Indralaya. The steps this research conducted were the isolation and purification of bacteria, selection test with sulphate content measurement at the Environment Agency of Palembang city, characterization morphology and phisiology of bacteria, and finally identification of bacteria used Bergey’s Manual of Determinative Bacteriology 8th and 9th, after that determined percentage similarity isolate with genus allegedly by Pearson correlation coefficient range. This research was obtained 5 isolates were able to reduce levels of sulphate of AAT there are P 2.1, P 2.2, P 3.1, P 4.1, and P 4.2. P 2.1 isolate have 74 % similarity with genus of Escherichia. P 2.2 isolate have 72 % similarity with genus of Desulfovibrio. P 3.1 isolate have 68 % similarity with genus of Listeria. P 4.1 isolate have 84 % similarity with genus of Enhydrobacter. P 4.2 isolate have 72 % similarity with genus of Hafnia.
