Abstract
Disposing of dyes into the water can disrupt aquatic ecosystems and cause disease for other living creaturs. Direct Red 80 dyes are often used in the production of Kain Jumputan. These dyes have a more concentrated color and difficult to degrade. Therefore we need a method of reducing dyes by not causing side effects, thats like using bacteria as a biodegradation agent. The bacteria were isolated from kain jumputan industry waste which was then regenerated in the laboratory. This research was conducted from January to March 2020. 8 types of indigenic bacteria were carried out selection process to get the type of indigenic bacteria that have the ability to biodegradate Direct Red 80 dyes. Bacterial of degrading agent is done by optimizing the concentration of dye, temperature and pH to increase the ability of biodegradation, biodegradation ability is calculated by the formulas the percentage of degradation power, then the biodegradation power is analyzed using Thin Layer Chromatography method. The highest biodegradation ability after optimization is at a concentration of 80 ppm dye, temperature 34ºC and pH 6 with a large percentage of 53.09% by A. jandaei BD 02 and 51.30% by P. guguanensis BD 14. Thin Layer Chromatography test was obtained comparison of Rf values, where A. jandaei get comparisons of Rf values; Rf 1 of 0.67 and Rf 2 of 0.62, wehereas P. guguanensis get comparisons of Rf values; Rf 1 of 0.55 and Rf of 2 0.49 with control has a value of 0.59.
