Abstract

The ethanol extract of petai pods contains of polyphenolic compounds, tannins, and flavonoids that are efficacious as antioxidants. Solid lipid nanoparticle (SLN) preparation formula aims to increase penetration through the stratum corneum. Preparations are formulated using solid lipid of cetyl alcohol, tween-80 surfactant, and propylene glycol co-surfactant. Variations in surfactant concentration, manufacture temperature, and stirring rate were used as factors with factorial design 23 in the Design Expert®10. Effect of factors composition and interaction was observed based on the results of the percentage efficiency entrapment (% EE), stability, pH, organoleptic, and hedonic. Results of the test analysis using Design Expert®10 yielded the optimum formula with the proportion of tween-80 0,704 mL, the temperature of manufacture of 60oC, and the speed of stirring 1000 rpm. Result of analysis % EE, particle size, polydispersity index (PDI), zeta potential, and viscosity are 88.808 ± 0.058; 393.8 nm; 0.421; -1.7 mV; and 2 cP. Qualitative testing of antioxidants using DPPH reagent showed SLN extract of ethanol pod of petai has positive antioxidant activity. Stability analysis using the thermodynamic method using temperature 4 ± 2oC and 40 ± 2oC for 6 cycles showed that optimum formula of SLN is more stable than pure extract. The percent diffusion measurements of the optimum formula and pure extract at 480 min were 9.830 ± 0.0114% and 4.484 ± 0.0219% indicating SLN penetration ability more bigger than ethanol extract of pod petai in carrier fluid. Compartemental analysis with WinSAAMTM software yielded the optimum formula following the compartment lag model with the correlation between Qo and Qc with p-value<0,05. The interaction studies of ethanol extracts pods petai and excipients using FTIR showed no chemical interactions.
