Abstract

Areca nut, purwoceng and ginseng jawa extracts have been known to have aphrodisiac effects or increased sexual arousal. The use of natural materials has limitations, including frequent failures in the clinical phase due to low bioavailability. Biaoavailability can be increased by shrinking the size of the material, so that nanoparticle technology can be used. The aims of this study were nanoparticle preparation and to determine and comparing the pharmacokinetic profile of the extract and nanoparticle preparations.. The preparation of extracted nanoparticles using biodegradable polymers was polylactic-co-glycolic-acid (PLGA) and as a stabilizer of polyvinyl alcohol (PVA). Followed by non-compartment pharmacokinetic studies using PKSolver add-in program. Areca nut, purwoceng and ginseng jawa extracts contain the majority of flavonoid compounds. Results from nanoparticle preparation on optimum formula 50 mg extract, 100 mg PLGA and 40 mg PVA with %EE 91.301%; PSA 584.9nm; PDI 0.364 and Zeta Potential -0.3mV. The morphology of the nanoparticles was spherical. The pharmacokinetic profiles of the extract and nanoparticles differed significantly (p<0.05) respectively, ka (min-1) 0.03; 0.08, Cmax (μg.ml-1) 3.14; 2.51, Tmax (min) 42.91; 34.81, AUC (μg.min.ml-1) 371.78; 487.64, Vd (ml) 7.07; 15.98, ke (min-1) 0.02; 0.007, T½ (min) 36.44; 108.23 and Cl (ml. min-1) 0.14; 0.103. Preparation of nanoparticles with polymerization techniques herb of areca nut, purwoceng and ginseng jawa extracts using PLGA and PVA are effective in enhancing pharmacokinetic profile. Both the increase of the absorption kinetics and the kinetics of the distribution as well as the slowing of the elimination kinetics so as to make effective use of them.
