Abstract
Ultrasonic-assisted extraction is an alternative extraction method for extracting natural compounds. Optimization of ultrasonic extraction bandotan leaf aims to determine the optimal solvent concentration and extraction time based on determination of total flavonoid content and IC50 antioxidant activity. The study was conducted by varying the solvent concentration (30, 70, 96%) and ultrasonic extraction time (20, 40, 80 min). Determination of the best extraction conditions is done using factorial design. Based on Design Expert 10® analysis, solvent concentration factor and ultrasonic extraction time have effect on total flavonoid content and antioxidant activity of bandotan leaf. The best ultrasonic exctraction condition are obtained at 70% ethanol concentration and 40 min extraction time selected based on the highest desirability value. Ultrasonic extraction using 70% ethanol for 40 minutes resulted 51.733 mg/g total flavonoid and 116.636 μg/mL IC50 antioxidant values.
