Abstract
Ethanol extract of Parkia speciosa Hassk. pods contains flavonoid, polyphenol, and tannin as antibacterial and antioxidant. The transfersome preparation was formulated using soy lecithin and tween-80 as factor with 32 factorial designs in Design Expert®10 program. The effect of factor compositions and interaction were observed based on the results of percent encapsulation efficiency (%EE), polydispersity index (PDI), pH, and percent decreasing of extract. Optimum formula was obtained with the concentration of lecithin and tween-80 of 0.85 g and 0.15 g resfectively with desirability 0.552. The resulting %EE, PDI, diameter, zeta potential and viscosity analysis of the optimum formula were 91.6884 ± 0.0261%, 495.6 nm, 0.484, -21.4 mV, and 3.1667 cP. After the stability assay of heating cooling using six cycles with defferent temperature of 4 and 40oC, the result was more stable than pure extracts. Diffusion analysis of the formula resulted into a higher diffuse percentage compared to the extract by itself which was 6.6253 compared to 1.8800%, this indicate that the transfersome has greater ability of penetration compared to the pure extract of petai pods. Compartement analysis using the WinSAAM® program followed the lag time model of 3 compartement with the correlation between Qc and Qo value of the greater than p- value of >0.5. Interaction analysis was conducted using the FTIR model using the placebo as the optimum formula, which the result indicates that is no chemical interaction between the formula as well as polymer compared to the placebo which showed that it was stable and it is applicable to the formulated into transfersome. Based on these result, the ethanol extract was suitable to made in transfersome because it had a higher diffuse percentage compared to the pure extract
