Abstract
Background: Effectiveness of antibacterial agent was assessed by the efficacy of antibacterial agent depressed bacterial growth. Natural antibacterial as alternative antibacterial agent which was considered safe, relatively has no side effects, inexpensive and common.Miana leaf as a plant that was known to have an antibacterial effects by its compounds such as flavonoid, alkaloid, tannin, and saponin. Application of antibacterial agent was needed to control oral bacterial growth like Streptococcus mutans as the main etiology of dental caries.Objective: The aim of this study was to determine antibacterial effectiveness of miana leaf extract againts the growth of Streptococcus mutans. Materials and Methods: Material which used in this study was miana leaves. This study was an experimental laboratory in vitro with post-test only control group design.The treatment group used miana leaf extract with concentration of 15,62 mg/ml, 31,25 mg/ml, 62,5 mg/ml, 125 mg/ml, and 250 mg/ml. Positive control used chlorhexidine 0,2% while negative control used dimethyl sulfoxide (DMSO). Inhibition zone tested using disk diffusion method, MIC and MBC tested using dilution method. Result: The average inhibition zone from miana leaf extract against Streptococcus mutans growth at concentration of 15,62 mg/ml was 3,80±0,20 mm, 62,5 mg/ml was 5,25±0,25 mm, 125 mg/ml was 7,50±0,30 mm, 250 mg/ml was 9,80±0,20 mm. Inhibition zone wasn’t formed at miana leaf extract concentration of 15,62 mg/ml and negative control group. MIC obtained at concentration of 62,5 mg/ml and MBC determined at concentration 125 mg/ml. Conclusion: Miana leaf extract effective as antibacterial against the growth of Streptococcus mutans.
