Abstract

A study of antidiabetic activity of ethanol extract of kirinyuh leaves (Chromolaena odorata (L.) R. M. King & H.Rob.) and histopathology of alistone-induced alloxan-induced wistar rats. Secondary metabolites of kirinyuh leaf were separated by maceration method with 96% ethanol solvent. Kirinyuh ethanol extracts contain flavonoids, alkaloids, tannins, phenolics, saponins, steroids, and triterpenoids. Ethanol extract kirinyuh leaf made with the level of dose variation that is 150, 300, and 600 mg/kgBW. As a positive control compound used insulin dose 1 IU/kgBW, negative control of suspension of Na-CMC 0.5%. Tests were performed by measuring fasting blood glucose levels of rats using GOD-PAP enzymatic method using DTN-410-K photometer on days 0, 10, 15, and 20. The mean percentage of decreased blood glucose levels for positive control group was 31.28%, and 3 groups of ethanol extract extract of leaf kirinyuh dose 150, 300, and 600 mg/kgBW respectively 27.29%, 34.68%, and 52.24%. This shows that the highest ethanol extract 600 mg/kgBW has the best antidiabetic activity compared with insulin and other dose with significant difference (p<0.05). Histopathologic preparations are prepared according to standard procedures with hematoxylin-eosin staining. The variables examined in this study are endemrin cell necrosis of Langerhans Islet in the pancreas. The histopathologic features showed necrosis of Langerhans islet endocrine cells in the negative control group and improvement in the positive control group and the three treatment groups. The best improvement was seen in the dose group of 600 mg/kgBW. This suggests that giving the highest doses of chloroethane ethanol extract of 600 mg/kgBW can lower blood glucose levels and improve the pancreas image that has been damaged better than other doses.
