Abstract

Perairan rawa dan kolam merupakan perairan yang menggenang (lentik) proses terbentuknya ada secara alamiah (rawa) sedangkan terbentuk secara buatan (kolam). Perairan rawa dan kolam mengandung keanekaragaman mikroorganisme salah satunya Cyanobacteria. Cyanobacteria disebut juga alga hijau biru yang hidup soliter dan berkoloni. Penelitian ini bertujuan untuk mengetahui jenis dari Cyanobacteria dengan identifikasi Cyanobacteria berasal kolam budidaya ikan perairan rawa di Indralaya. Penelitian ini telah dilakukan pada bulan Januari 2018 –Agustus 2018 di Indralaya. Analisis penelitian yaitu konsentrasi biomasa sel, kualitas air dan sekuen DNA. Berdasarkan morfologi diduga isolat kolam menyerupai Synechoccocus sp. dan isolat rawa menyerupai Microcystis sp. Amplikasi DNA Cyanobacteria menggunakan metode PCR dengan universal 16S rRNA 63F (Forward) dan 1387 R (Reverse) menghasilkan 1302-1307 bp. Analisis melalui BLAST (Basic local Aligment Search Tool- nucleotide) isolat kolam (Uncultured Synechoccocus sp. 91%) berasal dari Austalia sedangkan isolat rawa (Microcystis sp. 86%) berasal dari China.
