Abstract

Cocoa leaves contain flavonoids, alkaloids, and saponins which are thought to reduce uric acid levels. The purpose of this study was to determine the ability of the ethanol extract of cocoa leaves as anti-hyperuricemia in male white rats of Wistar strain induced by chicken liver juice and melinjo seeds. This study used male white rats Wistar strain which were divided into 6 groups, namely the positive group (allopurinol 90 mg/kgBW), the negative group (0.5% NaCMC ), and the ethanol extract test group of cocoa leaves with doses of 108, 216, 432, and 864 mg/kgBW. Uric acid data were analyzed using the ANOVA test to see differences in data at the time of observation. Measurement of uric acid levels was carried out by enzymatic methods using uric acid reagent TBHBA. Based on post hoc analysis (Duncan), there was no significant difference (p>0.05) between the EEDK treatment group and the positive control (allopurinol) because each extract had an effect on reducing uric acid levels. The results showed that the percentage reduction in uric acid levels in the groups with doses of 108, 216, 432, and 864 mg/kgBW were 29.23%, 37.95%, 52.02% and 78.51%. Based on the relationship of the percentage reduction in levels uric acid (% PKAU) to the extract dose, then the effective dose value of 50 (ED50) ethanol extract of cocoa leaves was 413.71 mg/kgBW. The total flavonoids contained in the ethanol extract of cocoa leaves were 96.50 mgQE/g. The ethanol extract of cocoa leaves has a significant effect on reducing uric acid levels and has potential as an anti-hyperuricemia.
