INSTAR III ABILITY OF EXTRACT AND FRACTION OF JAPANESE PEPAYA (Cnidoscolus aconitifolius (Mill) I.M. Johnst.) LEAVES ON MORTALITY OF LARVAS Aedes aegypti L INSTAR III. Nyamuk Ae. aegypti dapat menyebabkan penyakit salah satunya (demam berdarah dengue) DBD. Insektisida kimia lebih disukai masyarakat karena kerjanya cepat dan efektif, namun masyarakat tidak memikirkan dampak negative penggunaan insektisida kimia terhadap kesehatan manusia dan lingkungan. Oleh karena itu dibutuhkan alternatif lain sebagai pengganti penggunaan insektisida sintetis, seperti penggunaan insektisida nabati. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak dan fraksi daun pepaya jepang terhadap larva instar III Ae. aegypti dan mengetahui nilai LC50 ekstrak dan fraksi daun pepaya jepang terhadap larva instar III Ae. aegypti. Rancangan percobaan pada penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 7 konsentrasi dan 5 pengulangan. Prosedur pada penelitian ini yakni pertama mengetahui kemampuan ekstrak dan fraksi daun pepaya jepang (Cnidoscolus aconitifolius) dengan menguji ekstrak dan fraksi terhadap mortalitas larva instar III Aedes aegypti, diamati mortalitas dan kemudian dimasukkan dalam rumus % mortalitas. Kedua, mengetahui nilai LC50 ekstrak dan fraksi daun pepaya jepang dengan menganalisis data mortalitas yang sudah ada dengan menggunakan Analisis Probit. Hasil penelitian ini diketahui bahwa fraksi etil asetat memiliki kemampuan paling tinggi dalam membunuh larva uji dengan persentase kematin larva uji sebesar 100%. Nilai LC50 ekstrak methanol air daun pepaya jepang yang mampu membunuh 50% dari total larva uji adalah 2211,105 ppm, sedangkan nilai LC50 fraksi etil asetat adalah 2116,986 ppm. Daun pepaya jepang mengandung senyawa berdasarkan hasil KLT yaitu flavonoid, fenol, dan terpenoid. Kata Kunci : Kata kunci: Larvasida, Cnidoscolus aconitifolius, Aedes aegypti ABSTRACT : Mosquitoes are one of the vectors of diseases that can cause death to humans, including the Ae. aegypti. Ae. aegypti can cause disease, one of which is dengue hemorrhagic fever. Chemical insecticides are preferred by the community because they work quickly and effectively, but people do not think about the negative impact of using chemical insecticides on human health and the environment. Therefore, other alternatives are needed as a substitute for the use of synthetic insecticides, such as the use of vegetable insecticides. This study aims to determine the ability of extracts and fractions of Pepaya Jepang leaves against third instar larvae of Ae. aegypti and knowing the LC50 value of extract and fraction of Pepaya Jepang leaves against third instar larvae of Ae. aegypti. The experimental design in this study used a completely randomized design (CRD) with 7 concentrations and 5 repetitions. The procedure in this study was to first determine the ability of the extract and fraction of Pepaya Jepang leaves (Cnidoscolus aconitifolius) by testing the extract and fraction on the mortality of the third instar larvae of Ae. aegypti, observed mortality and then included it in the formula for % mortality. Second, knowing the LC50 value of extracts and fractions of Pepaya Jepang leaves by analyzing existing mortality data using Probit Analysis. The results of this study showed that the ethyl acetate fraction had the highest ability to kill test larvae with a 100% mortality percentage of test larvae. The LC50 value of methanol extract of Pepaya Jepang leaf water which was able to kill 50% of the total test larvae was 2211.105 ppm, while the LC50 value of the ethyl acetate fraction was 2116.986 ppm. Pepaya Jepang leaves contain compounds based on TLC results, namely flavonoids, phenols, and terpenoids. Keywords: Larvicides, Cnidoscolus aconitifolius, Aedes aegypti.
