Abstract

Bacillus sp. and Sterptomyces sp. from swamp so far relatively had not been able to be strored for too long, so that each time it will be used, it must be re-isolated. The solution which can be done is by stored in the appropriate media, so it can extend the stored time. The media that can be used are rice flour and tapioca which are expected to maintain viability of bacterial. Bacterial viability in quantity and quality can be known from bacterial population and patogen inhibition (A. hydrophila). The aim of this research are to know the viability and pathogen inhibition (A. hydrophila) of Bacillus sp. and Streptomyces sp. during the stored time in powder formulation. This research will use a completely randomized design with two factors and three replication. The first factor with two treatments are TB: formulation of rice flour 100 g, CMC 1 g, glucose 1 g and yeast 1 g; TT: tapioca formulation 100 g, CMC 1 g, glucose 1 g and yeast 1 g. The second factor with three treatments are I1: isolates Bacillus sp. 1010 CFU.ml-1; I2: Streptomyces sp. 1010 CFU.ml-1 ; I3: Bacillus sp. 5×109 CFU.ml-1 and Streptomyces sp. 5×109 CFU.ml-1. This research parameters are viability, total population, pH of media, moisture content and inhibition zone will be analyzed with analysis of variance and if the results are significantly different then it will be continued with LSD (Least Significance Different) 5%. The result of study showed that rice flour is better in maintaining viability of bacteria. The best treatment was isolate Streptomyces sp. with rice flour (TBI2), that be able to maintain viability 76,26 %, the highest total population 7,22 log cfu.g-1 and inhibition ability of Aeromonas hydrophila 6,67 mm.
