Abstract
Chitooligosaccharide is a chitosan derived compound resulting from the deacetylation of chitin which has great potential as a natural source of prebiotics due to its indigestible nature and beneficial effects on probiotic bacteria. This study aims to determine the effect of submicroscopic Chitooligosaccharide in stimulating the growth of Lactobacillus casei strain Shirota. Chitooligosaccharide submicroparticle preparation using sodium alginate as polymer and 20 µL CaCl2 0.018 M as crosslinker using ionic gelation method. The three formulas used variations in the weight of sodium alginate as much as 1.6; 3.2 and 4.8 mg. The best formula was found in the weight of sodium alginate 4.8 mg which had a percent encapsulation efficiency of 90.72% with a particle size characteristic of 556.8 nm; PDI value is 0.540 and zeta potential is +36.1 mV. The results of the bacterial growth activity test showed that Chitooligosaccharide submicro-particles could increase the growth of Lactobacilllus casei by 6.0 x 108 CFU/mL (13%) compared to Chitooligosaccharide without encapsulation of 5.3 x 108 CFU/mL. Chitooligosaccharide submicro particle stability test using heating-cooling method for 6 cycles with the results of the percent encapsulation efficiency (%EE) of 89.97; 70.56; 53.81; 30.45; 22.98 and 16.16% showed a decrease in the percentage of encapsulation efficiency in submicroparticle preparations.
