Abstract
It has been done the research of the Isolation and characterization of bacteria from tofu factory waste soil with dry heat method. In this research bacteria were isolated using soil extract medium and it was incubated by dry heat method at 50° C for 2 - 4 days. From the dry heat method result, four bacterial isolates Ri, R2, R3 dan R4 were able to grow at temperature of 50° C. The research continued to characterisize bacteria isolates from soil waste antibiotics test to determine the extracellular ability from bacteria isolates produce antibiotic metabolite against bacterial test Escherichia coli, Bacillus cereus, Staphylococcus aiireUSy, The inhibitory power of the test resulted that isolates R2 gave the largest inhibition zones on bacterial each for 6.6 mm, 8.6 mm and 3.0 mm for inhibition Escherichia coli, Bacillus cereus, Staphylococcus respectively, while isolates Rj gave inhibition 3,4 mm, 3,2 mm and 1,8 mm for inhibition Escherichia coli, Bacillus cereus, Staphylococcus respectively, and then R3 are 6,8 mm, 7,3 mm and 3,3 mm, and isolates for inhibition Escherichia coli, Bacillus cereus, Staphylococcus respectively, while R4 are 6,7 mm, 7,5 mm and 2,6 mm for inhibition Escherichia coli, Bacillus cereus, Staphylococcus respectively. Proteolytic activity from the test result only obtain two isolates with a clear each the value of R3 and R4 6.2 mm and 5.1 mm. Gram straining test showed that bacteria isolates Ry is a gram negative, R2 R3 and R4 are gram positive. Sugar cell test showed that isolates R\, R2, R3 and R4 has similar Rf with glucose and galactose. From the result of this characterization we can assume that isolate Ru R2, R3 and R4 are different bacteria.
