Abstract
Ultrasonic-assisted extraction and Microwave-Assisted Extraction is an alternative extraction method for extracting natural compounds. Optimization of ultrasonic extraction and Microwave Extractions of Cempedak leaf aims to determine the optimal solvent concentration and extraction time based on determination of total flavonoid content and IC50 antioxidant activity. The study was conducted by varying the solvent concentration (30, 70, 96%) and ultrasonic extraction time (20, 50, 80 minutes)on Ultrasonic Method and varying the microwave’s power (90 , 180, 360 Watt) and extraction time (10, 25, 40 minutes). Based on Design Expert 12® analysis, solvent concentration factor and ultrasonic extraction time have effect on total flavonoid content and antioxidant activity of cempedak leaf. Microwave’s power and extraction time have effect on total flavonoid content and antioxidant activity of cempedak leaf with microwave method. The best ultrasonic exctraction condition are obtained at 70% ethanol concentration and 80 min extraction time selected based on the highest desirability value. Ultrasonic extraction using 70% ethanol for 80 minutes resulted 548,930± 0,001247i mg/g total flavonoid content and 22,271±0,798332a µg/mL IC50 antioxidant values and the best microwave exctraction condition are obtained at 180 watt microwave’s power and 25 min extraction time selected based on the highest desirability value. Microwave extraction 180 watt microwave’s power for 25 minutes resulted 523,292±0,00047h mg/g total flavonoid and 32,64705±0,760948a µg/mL antioxidant values. Optimal conditions of two methods which 70% ethanol concetrations and 25 minutes extractions time used to maceration extractions and resulted 616,779 ±0,001247 mg/g total flavonoid content and ±19,011µg/mL IC50 antioxidant values.
