Abstract

Gambir water extract contain high catechin as polyphenol compounds that can be used as antioxidants. This antioxidant activity can be used in counteracting free radicals that cause Alzheimer's disease. Alzheimer's treatment is still limited in terms of effectivity because the target treatment is the brain. The intranasal preparation of gambir water extract micro particle is formulated to improve the effectiveness of anti Alzheimer's activity of gambir water extract with variation of chitosan concetration, crosslinker, and stirring speed using Box-Behnken design in Design Expert® version 10. Effect of chitosan concentration, type and concentration crosslinker, and stirring speed were observed against organoleptic response, percent EE, pH, stability, redispersion, time and sedimentation volume. The responses of optimum formulas have organoleptic value 5 and 4, percent EE 95.4570% and 95.9966%, pH 5.64 and 4.72, stability 64.6254 ppm and 76.3505 ppm, sedimentation time 679 seconds, redispersion 1, and sedimentation volumes 4 and 5. The optimum formula yields a particle size of 4559.9 and 2144.6 nm with a PDI value of 0.513 and 0.591 and zeta potential +9.5 mV. FTIR analysis results do not show the interaction between the polymer matrix with the extract so there is no presence of a new functional group. The diffused percent of microparticles of gambir water extract were 74.9201 and 71.1910% larger than the usual gambir water extract of 51.8368%. Compartmental analysis shows the extract and optimum formula follow lag time compartment. Based on this results analysis, extract micro particles from water gambir escould be use in intranasal drug delivery.
