Abstract
Alkaloids have been isolated from the bark of tembesu (Fragraea fragrans Roxb). This study complements the report on the alkaloid content of the tembesu bark. The antibacterial activity test was carried out against Staphylococcus aureus and Pseudomonas aeruginosa bacteria. Tembesu bark was extracted by maceration using diethyl ether and fractionated with ethyl acetate, separation and purification of alkaloid compounds using gravity column chromatography method. Structural elucidation was performed using LCMS/MS, 1H-NMR, 13C-NMR, and DEPT 135. The antibacterial activity test was carried out using the disc diffusion method and the minimum inhibitory concentration test using the liquid dilution method with the spectrophotometric test method. The concentration of bacteria used during the test was 1 x 108 CFU/mL. Alkaloid isolates from tembesu bark were obtained as much as 20.1 mg with a yield of 0.0017%. The data from the elucidation showed that the alkaloid isolate from the stem bark of tembesu contained an alkaloid compound in the form of 3,4-didehydrogentianin lactam which was still mixed with the compound (3R)-5'-methoxy-vestitol, ciwujiatone and ashantin. The results of the antibacterial activity test of tembesu stem bark isolates showed activity in the medium category and the minimum inhibitory concentration against Staphylococcus aureus and Pseudomonas aeruginosa at a concentration of 375 ppm which was categorized as moderate.
