Abstract
Background: Patients demonstrated response variability on drug delivery is still a problem in pharmacotherapy. One important factor that led to the phenomenon of occurrence is the difference between the ability of individuals to metabolize the drug. The enzyme glutathione S-transferase (GST) is enzyme that play a role in the process of phase II metabolism with one of the main components is glutathione S-transferase Pi-1 (GSTPI). Polymorphism in the coding gene for the enzyme GSTPl may affect the ability of GST enzyme to detoxify xenobiotics that have potency as carcinogenic. The purpose of the study is to indcntify the lle/05Val polymorphism ofGSTPl gene. Methods: This slu<ly is a labornlory research, is also an observasional descriptive study of 69 subjects from four major ethnics of Palembang population, are Melayu, China, Arab, and India ethnics. Identification of lle/05Val polymorphism GSTPI gene was done by using PCR amplification and RFLP (restriction fragment length polymorphism) technique using enzyme BsmAJ. Results: The genotype AA was found as much as 68,1%, 31,9% genotype AG, and not met genotype GG. Genotype AA as many as 36.2% in men and 31.9% in women. While the genotype AG was found as much as 20,3% in men and 11,6% in women. Genotype AA was found as much as 14,5% in the ethnic of Melayu, 27,5% in the ethnic China, 14,5% in the ethnic Arah. and 11.6% in the ethnic India. While genotype AG was found as much as 1l ,6o/o in the·ethnic of Melayu, 5,8% in the ethnic China, 11,6% in the ethnic Arab, and 2,9% in the ethnic India. Conclusion: The overview of genotype AA (wild type) were more common in four of mayor ethnics of Palembang population (68,1%).
