Abstract

Kulit buah pisang kepok (Musa balbisian C.) diketahui mengandung metabolit sekunder flavonoid yang memiliki efek antidiabetes. Tujuan dilakukan penelitan ini untuk mengetahui efektivitas penurunan kadar glukosa darah fraksi n-heksan kulit buah pisang . Penelitian ini dibagi menjadi 6 kelompok yaitu kelompok normal (suspense Na-CMC 0,5%), kontrol positif (metformin 9mg/kgBB), kontrol negatif (suspensi Na-CMC 0,5%) dan perlakuan fraksi n-heksan kulit buah pisang dengan variasi dosis 100, 200, dan 400 mg/kgBB. Tikus diinduksi aloksan dengan dosis 125 mg/kgBB hingga glukosa darah puasa ≥ 126 mg/dL, fraksi n-heksan kulit buah pisang diberikan selama 15 hari. Hasil penelitian menunjukkan persentase penurunan kadar glukosa darah fraksi n-heksan kulit buah pisang dosis 100, 200, dan 400 mg/KgBB berbeda signifikan dengan kontrol negatif (p<0,05), dengan persentase berturut – turut 44,59%; 49,43%; dan 58,07%. Dosis 400 mg/kgBB tidak berbeda secara signifikan dengan metformin (p>0,05) sedangkan dosis 100 dan 200 mg/kgBB berbeda signifikan dengan metformin (p<0,05). ED50 fraksi n-heksan kulit buah pisang adalah 217,67 mg/KgBB. Pengamatan histopatologi pankreas menunjukkan skor nekrosis sel β pakreas tikus pada kelompok dosis 400 mg/KgBB memiliki efek regenerasi sel terbaik dengan tingkat kerusakan 1-25% dibanding kelompok dosis 100 dan 200 mg/KgBB.
