Abstract
The use of usnic acid is still limited because it has low solubility, low bioavailability and lipophilic properties that make the ability of the usnic acid delivery system to penetrate the skin difficult to control. To overcome this problem, Solid Lipid Nanoparticles (SLN) were formulated which have a nanostructured lipid matrix so that they can provide more flexibility in modulating drug release. Preparation of SLN uses a combination of high-speed homogenization and ultrasonication. The SLN formula consisted of usnic acid, phospholipon 90G:cera flava (3:7), and tween 80 whose concentrations were varied to 0.3%; 0.6%, and 1%. The percentage of entrapment efficiency (%EE) becomes the parameter to determine the best formula where the %EE results in each formula are in succession 93%; 96% and 92% so that F2 with a concentration of Tween 80 0.6% became the best formula for further characterization of solubility, particle size, PI, zeta potential and stability tests. The resulting SLN particle size is 442.3 nm with a PI value of 0.282 and a zeta potential value of -15.2 mV. The stability test showed that SLN was stable in three storage conditions with release model following zero orde, activation energy 12,76 cal/mol, kinetic constant 0,11857 mg/days and shelf-life 40 days in storages condition 25oC/60%RH and in storage condition 5 oC 0,0826 mg/days in 58 days. The results of one-way ANOVA statistical analysis showed p value <0.05 where there was a significant effect of Tween 80 on the entrapment efficiency and SLN increased solubility compared to pure usnic acid. The results of the two-way ANOVA analysis for the test showed a p value <0.05 where there was a significant effect and there was an interaction between storage conditions and time in influencing the decrease in concentration. Keywords: Usnic Acid, Solid Lipid Nanoparticles, Phospholipon 90G, Cera Flava, Tween 80 Pemanfaatan asam usnat masih terbatas karena memiliki kelarutan rendah, bioavailabilitas yang rendah serta sifat lipofilik yang membuat kemampuan sistem penghantaran asam usnat menembus kulit yang sulit terkontrol. Untuk mengatasi hal tersebut diformulasikan Solid Lipid Nanoparticles (SLN) yang memiliki matriks lipid berstruktur nano sehingga dapat memberikan lebih banyak fleksibilitas dalam modulasi pelepasan obat. Pembuatan SLN menggunakan kombinasi homogenisasi kecepatan tinggi dan ultrasonikasi. Formula SLN terdiri dari asam usnat, phospholipon 90G:cera flava (3:7) dan tween 80 yang divariasikan konsentrasinya menjadi 0,3%; 0,6% dan 1%. Persen efisiensi penjerapan (%EE) menjadi parameter penentuan formula terbaik dimana hasil %EE pada masing�masing formula berturut-turut 93%; 96% dan 92% sehingga F2 dengan konsentrasi Tween 80 0,6% menjadi formula terbaik untuk selanjutnya dikarakterisasi kelarutan, ukuran partikel, PI, zeta potensial dan uji stabilitas. Ukuran partikel SLN yang dihasilkan 442,3 nm dengan nilai PI 0,282 dan nilai zeta potensial -15,2 mV. Pengujian stabilitas menunjukkan SLN stabil dalam tiga kondisi penyimpanan dengan model pelepasan mengikuti orde-0, energi aktivasi 12,76 cal/mol, konstanta kinetik 0,11857 mg/hari dan masa simpan selama 40 hari pada kondisi penyimpanan 25oC/60% RH dan pada kondisi penyimpanan 5oC 0,0826 mg/hari selama 58 hari. Hasil analisis statistik ANOVA satu arah menunjukan nilai p<0,05 dimana terdapat pengaruh Tween 80 yang signifikan terhadap efisiensi penjerapan dan SLN meningkatkan kelarutan dibandingkan asam usnat murni. Hasil analisis statistik ANOVA dua arah untuk uji stabilitas menunjukan nilai p<0,05 dimana terdapat pengaruh yang signifikan serta interaksi antara kondisi dan waktu penyimpanan dalam mempengaruhi penurunan kadar.
