Abstract

Pelangas (Aporosa octandra) is a plant belongs to Phyllanthaceae family which reported to have various traditional’s medicinal properties that have not been widely studied for their chemical content. The aims research are to know phenolic, flavonoid content and antioxidant activity of ethyl acetate extract from pelangas bark extract. The research was started with a phytochemical screening of the bark of the A.octandra plant. After that, Extraction by maseration method using a solvent on increasing polarity from n-heksane to ethyle acetate. The ethyl acetate extract was analyzed for total phenolic and flavonoid content using the photometric method and its antioxidant activity was determined using the DPPH (1,1-diphenyl-2- picrylhydrazyl) method. The results of phytochemical screening are positive for flavonoids, triterpenoids, steroids and saponins, while negative results for alkaloids. The total phenolic and flavonoid content of the ethyl acetate extract was 12.64 mgGAE/g and 7.33 mgQE/g. The antioxidant activity of ethyl acetate extract using the DPPH method obtained an IC50 value of 77.35 mg/L, which is included in the strong category of antioxidant. Pelangas (Aporosa octandra) termasuk dalam famili Phyllanthaceae dilaporkan memiliki berbagai khasiat obat tradisional yang belum banyak diteliti kandungan kimianya. Penelitian ini bertujuan untuk mengetahui total fenolik, flavonoid dan aktivitas antioksidan dari ekstrak etil asetat kulit batang pelangas. Penelitian diawali dengan melakukan skrining fitokimia terhadap kulit batang tumbuhan A. octandra. Selanjutnya dilakukan ekstraksi dengan menggunakan metode maserasi bertingkat dari n-heksana kemudian etil asetat. Selanjutnya dilakukan analisis kandungan total fenolik dan flavonoid menggunakan metode fotometri dan aktivitas antioksidannya ditentukan dengan menggunakan metode DPPH (1,1-diphenyl-2- picrylhydrazyl). Hasil penelitian menunjukkan skrining fitokimia diperoleh hasil positif mengandung senyawa kimia antara lain flavonoid, triterpenoid, steroid dan saponin, sedangkan alkaloid diperoleh hasil negatif. Kandungan total fenolik dan flavonoid pada ekstrak etil asetat sebesar 12,64 mgGAE/g dan 7,33 mgQE/g. Aktivitas antioksidan ekstrak etil asetat dengan menggunakan metode DPPH mendapatkan nilai IC50 sebesar 77,35 mg/L, yang termasuk kedalam antioksidan kategori kuat.
