Abstract

The purpose of this research were to determine physical, chemical and microbiological characteristics polar extracts of leaves and barks mangrove (Avicennia marina). The research was conducted at Agricultural Product Chemical and Agricultural Product Microbiology Laboratory, Agricultural Technology Department, Faculty of Agriculture, Sriwijaya University, from November 2018 to April 2019. The experiment used a Factorial Completely Randomized Design with two factors and each treatment was repeated three times. The first factor was type of plant part (Avicennia marina leaves and bark) and the second factor was maceration temperature using water solvent (40, 60, 80 and 100°C). The parameters were physical characteristic (color, lightness, chroma, hue), chemical characteristics (pH, total phenol, tannin level and antioxidant activity), microbiology characteristics (antibacterial activity). The results showed that the treatment of the part of the Avicennia marina mangrove plant types had significant effect on pH, tannin level, antioxidant activity and antibacterial activity of Staphylococcus aureus. The maceration temperature had significant effect on lightness, chroma, hue, pH, total phenol, tannin level, antioxidant activity and antibacterial activity of Staphylococcus aureus. The interaction of the two factors had significant effect on tannin level and antibacterial activity of Staphylococcus aureus. Based on this research the best treatment was treatment polar extracts of leaves mangrove; maceration temperature 80°C, It’s characteristic was: L 8,87%, C 2,63%, H 16,07°, pH 4,52, total phenol 0,41 mgTAE/g, tannin level 15,70 mg/l, IC50 2177,76 ppm and clear zone diameter formed on Staphylococcus aureus reaches 2,92 mm.
