Abstract

MUHAMMAD RIZAL ARDIANSYAH, Characteristic of collagenase Enzymes From Digstive Tract Bacteria of Snake Head Fish (Channa striata) (Supervised by ACE BAEHAKI). This study aims to determine the value of collagenase enzyme activity from the digestive tract bacteria of snakehead fish (Channa striata) by partially purifying the collagenase enzyme using ammonium sulfate and characterizing the collagenase enzyme o the effect of temperature. experimental laboratory method with extract testing carried out 3 (times) replicates, using collagenase enzyme activity testing. The data obtained were analyzed descriptively. The results showed that the activity value of the collagenase enzyme of snakehead fish with casein substrate was 1.729 U/ml which was higher than the collagen substrate from the digestive tract bacteria of snakehead fish which was 0.77 U/ml. The results showed that the intestinal bacterial supernatant of snakehead fish as a result of precipitation using ammonium sulfate increased up to a concentration of 60% and decreased at a concentration of 70-80%. The optimum concentration for the deposition of the collagenase enzyme supernatant from bacteria of 50% snakehead fish was 0.074 U/ml. The optimum environmental condition for collagenase enzyme activity is at 60°C with an enzyme activity of 0.36 U/ml, but the collagenase enzyme still has enzyme activity up to a temperature of 700C. Keywords: collagenase enzymes, enzyme activity, snakehead fish disgestive tract bacteria MUHAMMAD RIZAL ARDIANSYAH, Karakteristik Enzim Kolagenase dari Bakteri Saluran Pencernaan Ikan Gabus (Channa striata) (Dibimbing oleh ACE BAEHAKI) Penelitian ini bertujuan untuk mengetahui nilai aktivitas enzim kolagenase dari bakteri saluran pencernaan ikan gabus (Channa striata) dengan memurnikan enzim kolagenase menggunakan amonium sulfat secara parsial dan melakukan karakterisasi terhadap pengaruh suhu. Penelitian ini dilakukan menggunakan metode secara eksperimental laboratorium dengan pengujian kolagenase dilakukan sebanyak 3 (kali) ulangan, menggunakan pengujian aktivitas enzim kolagenase. Data yang didapatkan dianalisis secara deskriptif. Hasil penelitian menunjukkan nilai aktivitas enzim kolagenase ikan gabus dengan substrat kasein sebesar 1,729 U/ml lebih tinggi dari substrat kolagen dari bakteri saluran pencernaaan ikan gabus 0.77 U/ml. Hasil penelitian menunjukan bahwa Supernatan bakteri usus ikan gabus hasil pengendapan menggunakan amonium sulfat mengalami kenaikan sampai dengan konsentrasi 60% dan mengalami penurunan pada konsentrasi 70 – 80 %, Aktivitas optimum endapan kolagenase bakteri saluran pencernaan ikan gabus pada konsentrasi 60% sebesar 0.022 U/ ml dan konsentrasi optimum untuk pengendapan supernatan enzim kolagenase dari bakteri usus ikan gabus 50% sebesar 0,074 U/ml . Kondisi optimum lingkungan untuk aktivitas enzim kolagenase pada suhu 60°C dengan aktivitas enzim sebesar 0,36 U/ml akan tetapi enzim kolagenase masih mempunyai aktivitas enzim sampai suhu 700C .
