Abstract

Bacteria are microorganisms that can damage seafood and cause disease in humans. Borax and formalin are commonly used antibacterial agents, but have negative effects on health. Polyphenols in plant extracts are a source of natural antibacterial agents. This study aims The purpose of this study was to determine the antibacterial effectiveness of polyphenol compounds from ethanol extracts of nipah fruit fibers (Nypa fruticans) against Staphylococcus aureus and Pseudomonas aeruginosa. Polyphenol compounds were extracted by maceration method using 50% ethanol, the polyphenol extract content was determined by the Folin-Ciocalteu method, and antibacterial activity by the Kirby-Bauer disc diffusion method, each of which was carried out 3 times. The yield of the extract obtained was 12.32 ± 2.09%, with a polyphenol content of 14.585 mg GAE / g dry sample. Antibacterials produce an increase in sample concentration that is directly proportional to the inhibition of each bacteria. The diameter of the inhibition zone at a concentration of 1 mg/mL in Pseudomonas aeruginosa was 11.27 mm and Staphylococcus aureus was 9.13 mm. Based on these results, polyphenol compounds from watercress can be used as natural antibacterial agents. Bakteri merupakan mikroorganisme yang dapat merusak makanan laut dan menyebabkan penyakit pada manusia. Boraks dan formalin merupakan agen antibakteri yang umum digunakan, namun memiliki efek negatif terhadap kesehatan. Polifenol dalam ekstrak tumbuhan merupakan sumber agen antibakteri alami. Penelitian ini bertujuan Tujuan dari penelitian ini untuk mengetahui efektivitas antibakteri senyawa polifenol dari eksrak etanol serabut buah nipah (Nypa fruticans) terhadap Staphylococcus aureus dan Pseudomonas aeruginosa. Senyawa polifenol diekstrak dengan metode maserasi menggunakan etanol 50%, kadar polifenol ekstrak ditentukan dengan metode Folin–Ciocalteu, dan aktivitas antibakteri dengan metode difusi cakram Kirby-Bauer yang masing-masing dilakukan pengulangan sebanyak 3 kali. Rendemen ekstrak diperoleh sebesar 12,32±2,09%, dengan kadar polifenol sebesar 14,585 mg GAE/g sampel kering. Antibakteri menghasilkan peningkatan konsentrasi sampel berbanding lurus penghambatan masing-masing bakteri. Diameter zona hambat pada konsentrasi 1 mg/mL pada Pseudomonas aeruginosa yaitu 11,27 mm dan Staphylococcus aureus yaitu 9,13 mm. Berdasarkan hasil tersebut, senyawa polifenol dari selada air dapat dijadikan sebagai agen antibakteri alami.
