Abstract

Pain and inflammation are common health issues experienced by patients. Non-steroidal anti-inflammatory drugs (NSAIDs) are often used to treat these conditions, but they come with undesirable side effects. Traditional medicine using Gelam leaves (Melaleuca leucadendra L.) may offer a promising alternative with potential analgesic and anti-inflammatory properties. Secondary metabolites in Gelam leaves are believed to play a role in alleviating pain and inflammation by inhibiting prostaglandin production. This study aims to evaluate the effectiveness of Gelam leaf extract as an analgesic, indicated by reduced writhing behavior in mice following 0.6% acetic acid induction in the writhing test, and as an anti-inflammatory agent, demonstrated by reduced paw edema in mice following 1% carrageenan induction in the rat paw edema test. The study involved 24 male Wistar rats, divided into 6 groups: positive control (Diclofenac sodium 50 mg/kg), negative control (CMC 1%), ethanol extract, n-hexane fraction, ethyl acetate fraction, and ethanol-water fraction, all at a dose of 250 mg/kg. GC-MS analysis of the ethanol extract revealed two peaks: β-Caryophyllene at the first peak, and α-humulene, β-ocimene, and β-selinene at the second peak. The positive control group showed 72.44% analgesic and 73.44% anti-inflammatory activity. The best analgesic and anti-inflammatory effects were observed in the n-hexane fraction, with 69.23% analgesic activity and 57.85% anti-inflammatory inhibition, followed by the ethanol extract (64.70% analgesic, 49.56% anti-inflammatory), ethyl acetate (34.48% analgesic, 30.47% anti-inflammatory), and ethanol-water fraction (25.35% analgesic, 23.93% anti-inflammatory).
