Abstract
Excessive free radical production can lead to oxidative stress that damages DNA, proteins and lipids, triggering serious neurodegenerative and autoimmune diseases. Oxidative damage caused by free radicals can be prevented by the activity of antioxidant compounds. Based on previous research, papaya leaves, melinjo leaves, and pelawan leaves as natural antioxidants contain flavonoids and phenolic compounds with bioactivity mechanisms that reduce free radicals. The combination of these three plants has the potential to produce higher antioxidant activity because it can produce a synergistic effect in reducing stress due to free radicals. In this study, the fractions were combined with the aim of obtaining stronger antioxidant bioactivity than the single fraction. This process was carried out by combining the active fractions tested for antioxidants through the DPPH (1,1-diphenyl-2-picrylhydrazyl) method. The results showed the best combination with synergistic interactions in melinjo ethanol fraction (IC50 41,2 µg/mL) and pelawan (IC50 5,83 µg/mL) which resulted in an IC50 value of 3,87 µg/mL, melinjo ethanol (IC50 41,3 µg/mL) and papaya (IC50 106,73 µg/mL) which resulted in an IC50 value of 16, 02 µg/mL, ethyl acetate melinjo (IC50 101,22 µg/mL) and papaya (IC50 113,79 µg/mL) which produced an IC50 value of 21,74 µg/mL and a combination of n-hexane melinjo (IC50 157,16 µg/mL) with papaya (IC50 178,27 µg/mL) which produced an IC50 value of 120,54 µg/mL. Based on the test results, it can be seen that the combination of plants with synergistic effects showed stronger antioxidant activity than the antioxidant activity of plants produced singly.
