Abstract

Ethanol extract of Chinese Betel Herb (Peperomia Pellucida (L.) Kunth) is known to contain secondary metabolites, including flavonoid compounds that are thought to have potential antidepressant activity. This study aims to determine the effect of ethanol extract of Chinese betel herb as an antidepressant and determine the best concentration of ethanol extract of Chinese betel herb. Parameters observed included the length of time to fall, the number of line crossings, immobility time, weight change, the amount of sucrose consumption, and brain histopathology. Mice were divided into 6 groups: normal group (NaCMC 0.5%), negative group (NaCMC 0.5% + CUMS), positive group (fluoxetin 20 mg/kg BW + CUMS), and treatment groups I, II, and III (ethanol extract of Chinese betel herb at doses of 100, 200, and 400 mg/kg BW, respectively). Antidepressant testing of each group was carried out for 39 days using the forced swimming test, tail suspension test, traction test, locomotor, body weight measurement, sucrose preference, and brain histopathology observation. The results showed that ethanol extract of Chinese betel herb has antidepressant activity in mice induced by acute and chronic stress, compared to the negative control (p<0.05). In general, the test results showed an antidepressant effect of ethanol extract of Chinese betel herb against acute and chronic stress compared to the negative control.
