Abstract
Lichen has a secondary metabolite of usnic acid with antibacterial, antioxidant and anti-inflamatory pharmacological activities, however this compound is classified in Biopharmaceutics Classification System (BCS) class II with low solubility but high permeability so that it becomes an obstacle in topical formulation. This research aims to determine the optimum formula for usnic acid nanoemulsion with varying concentrations of peppermint oil (5%-10%) and Cremophor RH-40 (5%-10%) using the 22 factorial design method design method and testing the burn wound healing activity on wistar rats. The method of this research is making usnic acid nanoemulsion by spontaneous emulsification and testing the activity on healing burns where a hot plate is attached to create a wound. The optimum formula for usnic acid nanoemulsion with a combination of 5% peppermint and 10% Cremophor RH-40 based on research results was obtained with a desirability value of <1. The optimum formula was then evaluated and obtained a particle size of 166.73 ± 25.36 nm, a zeta potential of -22.73 ± 1.03 mV and a polydispersity index of 0.34 ± 0.07. Nanoemulsion shows good stability because it does not experience phase separation in the cycling test. The results of the burn wound healing test obtained the optimum formula was not significantly different from the positive control with a recovery percentage of 96,76±2,06 in 12 days. Usnic acid nanoemulsion has the potential to be developed as a wound healing drug. Keywords : Usnic acid, scabs, burns, nanoemulsion
