Abstract

Telah dilakukan penelitian mengenai aktivitas antihiperlipidemia fraksi n-heksana daun melinjo (Gnetum gnemon L.) terhadap tikus putih jantan galur Wistar yang diinduksi propiltiourasil. Tikus putih jantan galur Wistar dibagi menjadi enam kelompok yaitu normal, kontrol positif (simvastatin 0,987 mg/KgBB), kontrol negatif (NaCMC 0.5%), dan tiga kelompok uji dengan dosis 20 mg/KgBB, 40 mg/KgBB, dan 80 mg/KgBB. Pengukuran kadar trigliserida, dan kolesterol menggunakan metode enzimatik (GPO-PAP) dan (CHOD-PAP). Pengukuran kadar HDL menggunakan Cholesterol HDL Analysis Kit dengan alat fotometer, sedangkan kadar LDL digunakan perthitungan indirect. Penurunan kadar trigliserida, dan peningkatan HDL fraksi n-heksana dosis 20, 40, dan 80 mg/kgBB berbeda signifikan dengan simvastatin (p>0,05), sedangkan penurunan kadar kolesterol dan LDL fraksi n-heksana dosis 80 mg/kgBB tidak berbeda signifikan dengan simvastatin (p<0,05). Berdasarkan hubungan persentase penurunan parameter hiperlipidemia, maka dosis efektif (ED50) fraksi n-heksana daun melinjo sebesar 84,821 mg/kgBB. Pengamatan struktur histopatologi pada preparat penelitian ini meliputi nekrosis dan penimbunan/infitrasi lemak. Hasil pemeriksaan histopatologi organ hati tikus menunjukkan bahwa fraksi dosis 80 mg/kgBB sama seperti simvastatin dengan %nekrosis dan %degenerarasi lemak sebesar 10%. Hal ini menunjukkan bahwa fraksi n-heksana daun melinjo memiliki aktivitas sebagai antihiperlipidemia dan terdapat perbedaan signifikan dibandingkan dengan kelompok kontrol negatif (p<0,05).
